Table of Contents

Purpose

Previously, we almost did an exact replicate of our IC50-in-tubes assay (IC50 assay with 4 true biological replicates) , but altered the dilution factor and got different results. See the table at the bottom of this page for a rundown of what’s similar and different between experiments in the past.

The goal of this experiment is to:

• Do an in-plates experiment, while using our new 4X bench top shakers, that is as similar as we can get it to our first experiment: (Endpoint “IC50” assay)
• Try out the new (Endpoint IC50 Assay Protocol) using the new faster plate-shakers.

Experimental Overview

Monday (8/19): Starts overnight 1

Tuesday (8/20): Start salt plates rep 1.

Wednesday (8/21): Read OD600’s/ do data Analysis. Starts overnight 2.

Thursday (8/22): Starts salt plates rep 2.

Friday (8/23): Harley and Edward read OD600’s/Data Analysis

Experimental Notes

Start these strains in a 96wp with 300uL LB + Kan. Incubate overnight at 37C at 1000rpm.
